Laser-based measurements in cell biology.
In this chapter, we review the imaging techniques and methods of molecular interrogation made possible by integrating laser light sources with microscopy. We discuss the advantages of exciting fluorescence by laser illumination and review commonly used laser-based imaging techniques such as confocal, multiphoton, and total internal reflection microcopy. We also discuss emerging imaging modaliti...

New applications of laser microbeam irradiation to cell and developmental biology include a new instrument with a tunable wavelength (217- to 800-nanometer) laser microbeam and a wide range of energies and exposure durations (down to 25 X 10(-12) second). Laser microbeams can be used for microirradiation of selected nucleolar genetic regions and for laser microdissection of mitotic and cytoplas...
متن کاملLASER METHODS IN MEDICINE AND BIOLOGY Laser Microtools in Cell Biology and Molecular Medicine
Lasers for micromanipulation in cellular or subcellular dimensions have gained remarkable interest within the entire community of life science research and applications. In 1962, Bessis, for the first time, focused a ruby laser into a microscope to perform micromanipulation of cells under microscopic observation [1]. Today, there exist two principles of laser micromanipulation that depend on th...
متن کاملLaser trapping of deformable objects.
We report the trapping and manipulation of bubbles in viscous glass melts through the use of a laser. This phenomenon is observed in bubbles tens of micrometers in diameter under illumination by low numerical aperture (NA = 0.55). Once the bubble was centered on the optical axis, it was trapped and followed a lateral relocation of the laser beam. This phenomenon is explained by modifications of...
متن کاملLaser Trapping of Radioactive Atoms
Light forces acting on radioactive atoms confine them in very dense and cold samples achieving large phase space density. We describe the processes of radioactive atom production, reduction of their velocity and capture. The trapped atoms form excellent sources for the study of atomic parity non-conservation, β decay asymmetry, other weak force processes, and α decay asymmetry.
